چکیده

Neural stem cells (NSCs) offer the potential to replace lost tissue after nervous system injury. This study investigated whether grafts of NSCs (mouse clone C17.2) could also specifically support host axonal regeneration after spinal cord injury and sought to identify mechanisms underlying such growth. In vitro, prior to grafting, C17.2 NSCs were found for the first time to naturally constitutively secrete significant quantities of several neurotrophic factors by specific ELISA, including nerve growth factor, brain-derived neurotrophic factor, and glial cell line-derived neurotrophic factor. When grafted to cystic dorsal column lesions in the cervical spinal cord of adult rats, C17.2 NSCs supported extensive growth of host axons of known sensitivity to these growth factors when examined 2 weeks later. Quantitative real-time RT-PCR confirmed that grafted stem cells expressed neurotrophic factor genes in vivo. In addition, NSCs were genetically modified to produce neurotrophin-3, which significantly expanded NSC effects on host axons. Notably, overexpression of one growth factor had a reciprocal effect on expression of another factor. Thus, stem cells can promote host neural repair in part by secreting growth factors, and their regeneration-promoting activities can be modified by gene delivery.

Remyelination improvement after neurotrophic factors secreting cells transplantation in rat spinal cord injury

Objective(s): Neurotrophic factors secreting cells (NTS-SCs) may be a superior cell source for cell-based therapy in neurodegenerative diseases. NTS-SCs are able to secrete some neurotrophic Such as nerve growth factor and glia-derived neurotrophic factor. Our primary aim was to assess transplantation of neurotrophic factor secreting cells derived from human adipose-derived stem cells (hADSCs) ...

P 116: The Effect of Galectin-3 and Lanthionine Ketimine Ester in Neural Recovery after Spinal Cord Injury

Spinal cord injury (SCI) is a trauma that disturbs motor, sensitive and autonomic function and directly impacts the quality of life. After physical damage, releasing of pro-inflammatory proteins and cytokines occurs and with collaboration of immune system cells, an immune response begins in the brain tissue. The result of neuroinflammation is edema, apoptosis and release of axonal growth inhibi...

P144: Therapeutic Application of Mesenchymal Stem Cells in Spinal Cord Injury Treatment

Spinal cord injury (SCI) is a neurologic disorder that have a significant impact on quality of life, life expectancy, and economic burden. SCI leads to irreversible neuronal loss and ultimately leads to paralysis. Mesenchymal stem cells (MSCs) are a promising source for cellular therapy because they have possessed the capacity of self-renewal and differentiation to several distinct mesenchymal ...

Neurotrophic factors, gene therapy, and neural stem cells for spinal cord repair.

Several experimental strategies have been employed to minimize tissue damage and to enhance axonal growth and regeneration after spinal cord injury. The transplantation of suitable cell types to provide an axonal growth substrate and the application of growth factors have been shown to augment morphological and sometimes functional recovery. In this review we discuss the use of neural stem cell...

Minocycline Enhance Restorative Ability of Olfactory Ensheathing Cells by Upregulation of BDNF and GDNF Expression After Spinal Cord Injury

Purpose: Spinal cord injury is a global public health issue that results in extensive neuronal degeneration, axonal and myelin loss and severe functional deficits. Neurotrophic factors are potential treatment for reducing secondary damage, promoting axon growth, and are responsible for inducing myelination after injury. Olfactory ensheathing cells (OECs) and minocycline have been shown to promo...
